What is   Nepafenac  ? Nepafenac is a novel ocular non-steroidal antipyretic, analgesic and anti-inflammatory drug (NSAIDs). Nepafenac is used to treat eye pain and inflammation caused by cataract surgery. Compared with traditional NSAIDs, the chemical structure of Nepafenac is favorable for the drug to rapidly penetrate the cornea and distribute at its target site. Therefore, it is helpful to reduce the accumulation of drugs on the surface of the cornea, reduce the occurrence of complications on the surface of the eye, and has the advantages of strong penetration, strong targeting effect and small toxic and side effects.

 

Function of  Nepafenac 

Nepafenamine, when administered via the eye, rapidly passes through the cornea and is converted to amifenic acid (an NSAID) by hydrolases in the eye tissue; Amifenic acid plays an anti-inflammatory and analgesic role by inhibiting prostaglandin H synthetase (cyclooxygenase) and blocking the synthesis of prostaglandin. As is well known, prostaglandins are mediators that cause inflammation in the eye, leading to breakdown of the blood-aqueous barrier, vasodilation, increased vascular permeability and leukocyte chemotactic, etc. In addition, prostaglandins can also contract the iris sphincter through non-cholinergic mechanisms, leading to the constriction of the eyes during and after ophthalmic surgery. When administered via the eye, NSAIDs inhibit the synthesis of prostaglandins in the iris, ciliary body, and conjunctiva, thereby preventing eye inflammation and reducing associated pain.

Application of  Nepafenac 

Geldanamycin is ansamycin antibiotic isolated from Streptomyces hygroscopicus. It is a potent antitumour metabolite that acts by binding to the 90-kDa heat-shock protein (Hsp90) essential to maintain the conformation, stability, activity and cellular localisation of several key oncogenic proteins such as ERBB2, C-RAF, CDK4, AKT/PKB, steroid hormone receptors, mutant p53, HIF-1α, survivin and telomerase hTERT.
